





New Simultaneous UV-Visible Spectrophotometric Methods for Estimation of Ofloxacin and Ketorolac Tromethamine in Ophthalmic Dosage Form

New, simple, rapid and economic spectrophotometric methods have been developed for simultaneous estimation of Ofloxacin (OFLOX) and Ketorolac Tromethamine (KETO) in ophthalmic dosage form. For this, simultaneous Equation method (method A), Absorbance ratio method (Method B) and First order derivative spectroscopy (method C) were used. The method A involved measurement of absorbance at two wavelengths, 289 nm and 322 nm, max of OFLOX and KETO respectively. For method B, wavelengths 301 nm (isobestic point) and 289 nm (max of OFLOX) were used. In case of method C, wavelengths 262 nm (Zero crossing of OFLOX) and 274 nm (Zero crossing of KETO) were used for determination of KETO and OFLOX respectively. Beer's law obeyed in concentration range of 1.5-10.5 μg/ ml, and 2.5-17.5 μg/ ml for OFLOX and KETO respectively by all the methods. The proposed methods are recommended for routine analysis since they are rapid, simple, accurate and specific. Methods developed also showed no interference from Bezalkonium Chloride which is used as a preservative in the formulation. These methods were validated for linearity, precision and accuracy as per ICH guidelines.
